君實生物(688180.SH)：簽署關於分子內二硫鍵IL-2藥物之技術許可合同
格隆匯 8 月 29日丨君實生物(688180.SH)公佈，近日，公司與北京志道生物科技有限公司(“志道生物”)簽署了《關於分子內二硫鍵IL-2藥物之技術許可合同》，公司將獲得授權在全球範圍內對IL-2藥物(項目代號：LTC002)進行臨牀前開發、臨牀研究及商業化並使用相關專利技術的獨佔許可。
財務條款，1、首付款：公司將向志道生物支付2100萬人民幣首付款。
2、里程碑付款及技術服務費：公司將根據研發、上市及銷售情況向志道生物支付累計不超過9.38億人民幣的里程碑款及技術服務費。
3、銷售提成：自LTC002實現商業化銷售開始，公司將按照約定比例向志道生物支付淨銷售收入6-8%的銷售提成。
白介素-2(IL-2)於1976年被發現，當時被稱為T細胞生長因子(TCGF)，是一種在維持T淋巴細胞和NK細胞的正常功能中起着重要作用的球狀糖蛋白。IL-2主要由活化的T細胞生成，它能促進T細胞的增殖和分化，維持T細胞活性；刺激天然殺傷(NK)細胞的生成、增殖和活化，並誘導細胞毒性T淋巴細胞(CTL)的生成以及誘導和激活淋巴因子激活的殺細胞(LAK)及腫瘤浸潤淋巴細胞；因此IL-2有很好的抗病毒、抗癌作用和廣泛的臨牀應用潛力。但是，目前已上市的野生型IL-2產品在低劑量的條件下，會優先和Treg細胞表面上的高親和力受體結合，產生免疫抑制，達不到治療的效果。高劑量的IL-2會通過激活大量的效應T細胞從而中和Treg激活帶來的免疫抑制，同時也會出現更多的毒副作用，以及細胞凋亡。志道生物採取了一種獨特創新的方式消除與IL2Rα受體的結合，利用在IL-2內部引入額外的二硫鍵，其不僅可以使得IL-2從結構上更加穩定，而且還可以形成屏障，使IL-2分子以最小的改變達到破壞與α受體的結合平面。突變分子無法與體內的內源性α受體結合，但是可以與β和γ受體亞基結合。因此降低或消除IL-2與α受體亞基的相互作用，可能是治療有效性並減少腫瘤患者治療副作用的一個重要方面。目前項目處於臨牀前開發階段。
