麗珠醫藥(01513.HK)：V-01序貫加強對於Omicron感染導致的COVID-19可產生良好保護力
格隆匯2月16日丨麗珠醫藥(01513.HK)公吿，近日，公司控股附屬公司珠海市麗珠單抗生物技術有限公司與中國科學院生物物理研究所合作研發的“重組新型冠狀病毒融合蛋白疫苗”（「V-01」）已完成序貫加強Ⅲ期臨牀試驗的中期主資料分析，並獲得了關鍵性數據。
V-01自2020年7月開始立項研發，是麗珠單抗與中科院合作研發的具有自主知識產權的創新型新冠重組蛋白疫苗。V-01於2021年年初獲中國國家藥品監督管理局（國家藥監局）批准進行臨牀試驗，適應症為預防新型冠狀病毒感染所致疾病（COVID-19）。V-01分子由干擾素、Pan表位、RBD二聚體、Fc融合蛋白四部分組成。由於抗原結構中融合了作為生物佐劑的人源干擾素，顯着增強病毒中和抗體水準，併產生有效的細胞應答。V-01兩針接種的II期臨牀試驗資料已於2021年7月在《中華醫學雜誌》發佈，試驗結果顯示該疫苗具有優良的免疫原性和安全性，尤其在老年組中，疫苗相關不良事件的總體發生率低於相應的成人組。
麗珠單抗於2021年10月首先取得巴基斯坦藥品監督管理局關於“評價重組新型冠狀病毒融合蛋白疫苗（V-01）的序貫加強在完成2劑滅活疫苗接種的18週歲及以上健康成年人中的保護效力、安全性和免疫原性的國際多中心、隨機、雙盲、安慰劑對照的Ⅲ期臨牀試驗”的批准，隨後又取得了馬來西亞關於序貫加強方案的臨牀試驗批准，加速開展研究。
截至目前，麗珠單抗在V-01項目累計直接投入的研發費用約為人民幣50,940.09萬元（含I/II期臨牀試驗、序貫加強及基礎免疫III期臨牀試驗相關費用等）。
本次序貫加強方案為二針滅活疫苗基礎上序貫加強設計，採用相對保護力強優效標準，與傳統空白安慰劑方案對照不同。V-01在巴基斯坦和馬來西亞開展序貫加強的III期臨牀試驗，計劃在完成2劑滅活疫苗接種3~6個月的18週歲及以上健康成年人共計10,722人中按1:1注射V-01加強針或空白安慰劑。截止到本次關鍵性數據分析日，實際入組10,241人，共監測到接種後的主要終點病例數110例（方案要求至少103例主要終點病例時進行分析）。
根據現階段分析的試驗結果顯示：V-01序貫加強後與兩針滅活疫苗對比，V-01序貫加強組和兩針滅活疫苗組人年發病率分別為6.73%和12.80%，具有顯著性差異（P=0.0012）；V-01序貫加強後的絕對保護力為61.35%，具有顯着強優效性。V-01序貫加強絕對保護力已滿足世界衞生組織的標準。本次試驗全部有效送檢標本中60個的新冠病毒基因分型一代測序均為Omicron（其餘仍在進行二代測序），表明V-01序貫加強對於Omicron感染導致的COVID-19可產生良好的保護力。同時，未發現有值得關注的安全性問題。
據LSHTM跟蹤器信息，截至2022年2月7日，全球共有121款新冠疫苗產品處於臨牀試驗階段，其中有27款產品已獲批上市或緊急使用授權。國內獲得附條件批准上市4款（無重組蛋白技術路線產品），獲得緊急使用批准7款（其中重組蛋白技術路線1款），共21款處在臨牀試驗階段（重組蛋白技術路線8款）。
