海思科(002653.SZ)：創新藥HSK16149膠囊Ⅱ/Ⅲ期臨牀試驗獲得倫理批准
格隆匯9月15日丨海思科(002653.SZ)公佈，公司於2020年8月向北京大學第一醫院醫學倫理委員會遞交了關於“一項採用適應性設計評估HSK16149膠囊在中國糖尿病周圍神經痛患者中有效性和安全性的多中心、隨機、雙盲雙模擬、安慰劑和普瑞巴林膠囊對照、13周的II/III期研究”的臨牀相關資料，並於近日獲得倫理批件，可開始實施II/III期臨牀研究。
HSK16149由公司自主研發，公司擁有HSK16149的專利在中國境內的獨佔實施許可權，目前擬開發治療糖尿病周圍神經痛及帶狀皰疹後神經痛的適應症。按我國藥品註冊管理辦法規定，其藥品註冊分類為化藥1類。
HSK16149於2018年11月9日獲得國家藥品監督管理局的《臨牀試驗通知書》；分別於2018年12月-2019年6月、2019年7月-2020年1月在上海市徐彙區中心醫院開展了2項I期臨牀試驗，2020年7月依據前期已完成的I期臨牀試驗數據同國家藥品監督管理局藥品審評中心溝通，公司做出本項目進入II/III期臨牀試驗的決定。近期已獲得北京大學第一醫院的倫理批件，後期即將開展篩選入組。
II/III期臨牀試驗相關情況如下：
本研究的組長單位是北京大學第一醫院，由郭曉蕙教授擔任本研究的主要研究者(PI)，全國40家研究中心將加入HSK16149的II/III期臨牀研究。
試驗名稱：一項採用適應性設計評估HSK16149膠囊在中國糖尿病周圍神經痛患者中有效性和安全性的多中心、隨機、雙盲雙模擬、安慰劑和普瑞巴林膠囊對照、13周的II/III期研究。
試驗設計：本研究為一項II/III期適應性設計的多中心、隨機、雙盲、安慰劑和普瑞巴林膠囊對照的試驗，計劃納入大約680例符合入組條件受試者，各中心競爭入組。
試驗目的：評估不同給藥劑量的HSK16149治療糖尿病周圍神經痛(DPNP)的有效性和安全性。
疼痛是DPNP常見的臨牀症狀，約見於50%左右的糖尿病和約13%的糖耐量受損的患者，嚴重影響其正常生理和精神狀態，出現睡眠障礙、營養失調、運動受限、情感障礙，從而降低生活質量和工作能力。DPNP為排他性診斷，在糖尿病或糖尿病前期基礎上，根據臨牀表現、神經系統查體及神經電生理檢查證實存在周圍神經病變。多項流行病學研究顯示，國外DPNP總體患病率在10～26%，美國神經病學學會(AAN)報道約16%的糖尿病患者發生神經痛，其中39%患者未得到治療。儘管國內尚無針對DPNP大規模的流行病學調查，基於中國龐大的糖尿病患者基礎人羣，預計DPNP患病率較高，嚴重影響着廣大糖尿病患者的生活質量。儘管國內外指南推薦多種鎮痛藥物選擇，DPNP患者對鎮痛治療的反應程度不盡相同，無論對單藥或聯合治療，部分患者疼痛控制均不理想，較大劑量用藥也帶來安全性問題。同時，同類藥物普瑞巴林DPNP適應症及邁瑞巴林在中國均未批准上市。因此，亟需研發新型藥物為DPNP的治療提供安全有效的新選擇。
