基石藥業-B(02616.HK)CS1001聯合拜耳公司瑞戈非尼臨牀研究完成首例患者給藥
格隆匯1月23日丨基石藥業-B(02616.HK)發佈公告，公司在研PD-L1抑制劑CS1001與Bayer Healthcare LLC(拜耳公司)開發的口服多激酶抑制劑瑞戈非尼的聯合用藥研究已在澳大利亞完成首例患者給藥。這是基石藥業和拜耳公司合作的首個全球概念驗證性臨牀研究，旨在探索CS1001聯合瑞戈非尼的推薦劑量，以及評估包括胃癌在內的多種癌症治療的安全性、耐受性、藥代動力學和抗腫瘤療效。
CS1001是由基石藥業自主研發的在研抗PD-L1單克隆抗體。目前，CS1001正在進行多項臨牀試驗，除了一項美國橋接性I期試驗外，在中國，CS1001正針對多個癌種開展一項多臂Ib期試驗，兩項註冊性II期試驗和四項III期試驗。根據第二十二屆全國臨牀腫瘤學大會上公佈的研究資料，在Ia期和Ib期研究中，CS1001已在多個癌種中顯示出良好的抗腫瘤活性和耐受性。
瑞戈非尼是由拜耳公司開發的以VEGFR、FGFR、CSF1R等為作用靶點的多激酶小分子抑制劑口服藥物。目前瑞戈非尼已被包括中國在內的全世界90餘個國家批准用於二線及以上晚期結直腸癌、晚期胃腸道間質瘤及晚期肝細胞癌。
2019年5月，基石藥業與拜耳公司達成一項全球臨牀合作研究，基石藥業作為CS1001聯合瑞戈非尼臨牀研究的發起者，拜耳公司將提供試驗所需的瑞戈非尼。
多項研究表明，激酶抑制劑單藥或抗PD-1/PD-L1抗體單藥在多個瘤種治療中均具有療效。同時，臨牀前研究表明，如瑞戈非尼這樣的靶向藥物可通過改變一些實體瘤的免疫微環境來增強免疫檢查點抑制劑的療效。因此，兩種作用機制聯合可能對腫瘤消退發揮協同作用。
基石藥業董事長、執行董事兼首席執行官江寧軍博士表示：基石藥業與拜耳公司共同開展的這項臨牀試驗是公司全球戰略的重要一步。很高興看到CS1001和瑞戈非尼的聯合治療研究實現了首例患者給藥。公司希望這次聯合探索不僅將能補充公司的產品管線，還有望讓更多缺乏有效治療方案的腫瘤患者獲益。
基石藥業首席轉化醫學官謝毅釗博士表示:臨牀前動物疾病模型上開展的研究表明，CS1001和瑞戈非尼聯合能夠增強抗腫瘤作用，這為公司開展此項臨牀研究提供了資料支援。同時，目前國際上針對晚期或轉移性實體瘤也正在開展多項此類聯合療法，並已取得部分良好資料，公司對CS1001和瑞戈非尼的臨牀試驗結果充滿期待。
據悉，瑞戈非尼是一種口服的多激酶抑制劑，可阻斷涉及腫瘤血管生成(VEGFR1,-2,-3,TIE2)、腫瘤增殖(KIT、RET、RAF-1、BRAF)、腫瘤轉移(VEGFR3、PDGFR、FGFR)以及腫瘤免疫(CSF1R)的多種蛋白激酶。瑞戈非尼是由拜耳公司開發的一種化合物。2011年，拜耳公司與生技製藥公司Onyx Pharmaceuticals,Inc.達成協議。按照協定，Onyx將根據瑞戈非尼在腫瘤領域全球淨銷售額獲得專利權使用費。目前，瑞戈非尼已經在90餘個國家獲得了批准，用於轉移性結直腸癌、轉移性胃腸道間質瘤和二線晚期肝細胞癌的治療，該藥批准的商品名為拜萬戈(Stivarga?)，批准的國家包括美國、歐盟各國、中國和日本。
