恆瑞醫藥(600276.SH)披露藥品氟唑帕利膠囊臨牀試驗進展
格隆匯 9 月 28日丨恆瑞醫藥(600276.SH)公佈，近日，公司PARP抑制劑氟唑帕利膠囊對比安慰劑用於複發性卵巢癌維持治療的隨機、雙盲、對照、多中心III期臨牀研究，由獨立數據監查委員會(IDMC)判定主要研究終點的期中分析結果達到方案預設的優效標準，研究結果表明，氟唑帕利單藥用於複發性卵巢癌(包括輸卵管癌、原發性腹膜癌)含鉑治療達到完全緩解或部分緩解後的維持治療，可顯著延長患者的無進展生存期。公司將於近期向國家藥品監督管理局藥品審評中心遞交上市前的溝通交流申請。
此研究是一項評估氟唑帕利膠囊對比安慰劑用於複發性卵巢癌(包括輸卵管癌、原發性腹膜癌)含鉑治療後維持治療的隨機、雙盲、安慰劑對照、多中心III期臨牀研究(NCT03863860)，由中國醫學科學院腫瘤醫院吳令英教授擔任主要研究者，全國35家中心共同參與。研究的主要終點為由盲態獨立影像評審委員會(BIRC)根據RECISTv1.1標準評估的無進展生存(PFS)，次要研究終點包括研究者評估的PFS、無化療間期(CFI)、總生存期(OS)和安全性等。
研究共入組252例受試者，按照2:1隨機入組，分別接受氟唑帕利膠囊或氟唑帕利模擬膠囊口服，治療直至疾病進展、毒性不可耐受或其他需要終止治療的情況。近日，由獨立數據監查委員會(IDMC)判定主要研究終點的期中分析結果達到方案預設的優效標準，研究結果表明，氟唑帕利單藥用於複發性卵巢癌(包括輸卵管癌、原發性腹膜癌)含鉑治療達到完全緩解或部分緩解後的維持治療，可顯著延長患者的無進展生存期。
根據我國藥品註冊相關的法律法規要求，藥物需完成臨牀研究並經國家藥品監督管理局審評、審批通過後方可生產上市。公司將按國家有關規定及時對項目後續進展情況履行信息披露義務。
氟唑帕利單藥治療BRCA1/2突變的復發卵巢癌的新藥上市申請於2019年獲得國家藥品監督管理局受理，並被國家藥品監督管理局藥品審評中心納入優先審評。除本次獲得陽性結果的FZPL-III-301-OC研究外，氟唑帕利單藥或聯合阿帕替尼治療卵巢癌、乳腺癌、胰腺癌已處於III期臨牀研究階段。氟唑帕利另有多種聯合治療方案，包括與阿比特龍聯合、與抗PD-L1抗體SHR-1316及替莫唑胺聯合治療多種實體腫瘤已處於臨牀開發階段。
氟唑帕利是一種聚腺苷二磷酸核糖聚合酶(poly(ADP-ribose)polymerase,PARP)抑制劑，可特異性殺傷BRCA突變的腫瘤細胞。經查詢，氟唑帕利目前國外有同類產品奧拉帕利(商品名Lynparza)、盧卡帕利(商品名Rubraca)、尼拉帕利(商品名Zejula)和他拉唑帕利(商品名Talzenna)於美國獲批上市銷售，奧拉帕利片於2018年8月在中國獲批上市，商品名為利普卓。國內再鼎醫藥的甲苯磺酸尼拉帕利膠囊(商品名則樂)於2019年12月在中國獲批上市，用於鉑敏感的複發性上皮性卵巢癌、輸卵管癌或原發性腹膜癌成人患者在含鉑化療達到完全緩解或部分緩解後的維持治療。經查詢，2019年奧拉帕利(商品名Lynparza)、盧卡帕利(商品名Rubraca)、尼拉帕利(商品名Zejula)和他拉唑帕利(商品名Talzenna)全球銷售額約為13.57億美元，美國銷售額約6.74億美元。截至目前，該產品累計已投入研發費用約27131萬元。
