衝擊一線治療！百悦澤®針對初治慢淋的全球3期研究達到主要終點
格隆匯7月30日丨2021年7月30日，百濟神州(06160.HK)宣佈，BTK抑制劑百悦澤®(澤布替尼)一項全球性3期SEQUOIA試驗的期中分析取得了積極的主要結果，研究達到主要終點。數據顯示，相比化學免疫治療，百悦澤®顯著延長了初治的慢性淋巴細胞白血病(CLL)患者的無進展生存期，並展示了良好的安全性與耐受性。
SEQUOIA是繼全球3期頭對頭ALPINE研究後，百悦澤®第二項在CLL中取得積極結果的全球性3期試驗，更是百悦澤®衝擊CLL一線治療，證實其治療地位的重要一役。
慢性淋巴細胞白血病(CLL)是成人中最常見的一種白血病，在2017年，全球約有114,000例新發的CLL病例。SEQUOIA是一項隨機、多中心、全球性的3期試驗(NCT03336333)，入組患者超過580例，旨在評價百悦澤®與苯達莫司汀聯合利妥昔單抗(B+R)用於治療初治CLL或小淋巴細胞淋巴瘤(SLL)患者的有效性和安全性。該試驗由三個隊列組成，而此次期中分析，對比了百悦澤®與B+R，用於治療未顯示17p13.1染色體缺失(del[17p])的初治CLL或SLL患者的數據。
中位隨訪時間為25.8個月時，百悦澤®相較B+R在無進展生存期(PFS)方面顯示出具有高度統計學顯著性的改善，因此SEQUOIA試驗達到其主要終點，即基於獨立審查委員會(IRC)評估結果確定的PFS。
此外，SEQUOIA試驗表明，作為一項次要終點，基於研究者評估的PFS也顯示了具有統計學顯著性的改善。同時，百悦澤®的耐受性總體良好，與其已知的安全性特徵一致。
百濟神州血液學首席醫學官黃蔚娟博士表示：“綜合三項全球性臨牀研究SEQUOIA、ALPINE、205試驗和AU-003試驗的數據，證實了我們對百悦澤®的信心，我們相信它能夠改善數十萬CLL患者的治療結果。我們很欣慰地在SEQUOIA試驗的期中分析中看到，百悦澤®顯著延長了初治CLL患者的無進展生存期，同時其顯示的安全性特徵與其在全球開發項目中觀察到的特徵一致，迄今為止，已有超過2,300例患者接受了百悦澤®治療。”
百濟神州表示，將與全球藥監機構就下一步計劃展開溝通，並在即將召開的重要醫學會議上公佈這些數據。
百悦澤®是一款由百濟神州科學家自主研發的BTK小分子抑制劑，目前正在全球進行廣泛的臨牀試驗項目，作為單藥和與其他療法進行聯合用藥治療多種B細胞惡性腫瘤。由於新的BTK會在人體內不斷合成，百悦澤®的設計通過優化生物利用度、半衰期和選擇性，實現對BTK蛋白完全、持久的抑制。憑藉與其他獲批BTK抑制劑存在差異化的藥代動力學，百悦澤®能在多個疾病相關組織中抑制惡性B細胞增殖。
百悦澤®已在美國、中國、阿聯酋、加拿大、以色列等國家或地區中獲批多項適應症，並先後在全球針對8項適應症開展了近30項臨牀試驗。目前，共有30多項百悦澤®針對多項適應症的相關上市申請完成遞交工作，覆蓋歐盟以及其他20多個國家或地區。在中國，百悦澤®已經被納入國家醫保目錄，並於2021年3月1日生效，以更親民的價格，惠及我國本土患者。
